Lipid-based nanocarriers, particularly lipid nanoparticles (LNPs), have transformed the landscape of nucleic acid delivery, finding success in mRNA vaccines and gene therapies. However, these delivery systems remain constrained by their inherent liver tropism, limiting broader therapeutic applications. Recent advances in modular peptide dendrimer lipid hybrids and artificial intelligence (AI)-driven screening techniques are now reshaping the field, offering greater precision and efficiency in targeting delivery systems beyond the liver.
