Advanced analytical strategies for AAV vector characterization using dPCR and long-read sequencing

Live30 webinars pack the latest innovations and applications into a data-rich 30-minute session.

Using digital PCR (dPCR) alongside long-read sequencing provides complementary insights into genome integrity and impurity profiles, giving you more reliable data for confident decision-making. This webinar will highlight how to adopt optimized workflows and standardized extraction approaches to streamline analysis, reduce variability and strengthen regulatory preparedness.

The session will present benchmarking data demonstrating how standardized recombinant AAV (rAAV) DNA extraction method minimizes sample bias and improves reproducibility compared with conventional lysis methods. It will also explore how dPCR enables the detection of encapsidated plasmid and host cell DNA contaminants, as well as the ability to analyze multiple regions of the AAV genome within a single reaction, enabling a clear distinction between full-length and truncated genomes.

The session will be led by Jens Stolte, Senior Research Engineer at New Biologix, who brings over 25 years of genomics and sequencing experience from leading global institutions. Real-world examples will highlight the trade-offs between sequencing- and dPCR-based integrity testing and provide guidance on selecting the most effective approach for your workflows. As an attendee, you will gain practical strategies to help optimize your analytical approaches, validate your processes and adopt tools that drive efficiency and consistency in AAV vector development.

Attend this webinar to:

  • Learn best practices for applying dPCR in AAV quality control, including dilution strategies to extend dynamic range and improve quantification accuracy
  • Discover how dPCR can be used to target multiple regions of the AAV genome in a single reaction, enabling a clear assessment of genome integrity and distinguishing between full-length and truncated genomes
  • Gain insights from benchmarking data showing how standardized DNA extraction improves reproducibility and reduces sample bias compared with conventional methods
  • Explore practical guidance on selecting and combining dPCR and long-read sequencing to strengthen AAV analytics and meet regulatory expectations

Jens Stolte
Senior R&D Engineer at New Biologix

Jens Stolte began his career over 25 years ago, as genomics took off with the completion of the Human Genome Project and the rise of microarray technology. He has since worked across academia and industry at leading research institutions worldwide, with contributions ranging from infectious disease genomics in Ghana to pioneering next-generation sequencing at EMBL Heidelberg. At EMBL, he also taught international qPCR courses for several years before moving on to cancer research at the EMBL Mouse Biology Outstation in Italy. He later joined Japan’s RIKEN Institute, where he advanced genomics technologies with a focus on long non-coding RNAs. In Switzerland, Jens expanded his expertise into epigenetics during his time at Nestlé Research, where he implemented Nanopore sequencing and bridged fundamental science with translational applications. He now serves as a Senior Research Engineer at New Biologix, where he develops technologies for AAV analytics and cell engineering, continuing his long-standing commitment to innovation in genomics.
